The Principle of Stable Cells Generation

The foreign DNA/shRNA is cloned into an expression vector with containing one resistance gene. The recombinant vector is then introduced into the host cell and integrated into its chromosome, so that the foreign DNA/shRNA can be transmitted to daughter cells upon cell division. The stable cell line are selected with the resistant gene on plasmid. The most commonly used selection antibiotics for stable cell transfection are neomycin, blasticidin and puromycin.
